AnotationThis work will focus on the development of analytical method for determination of selected drugs in human serum for the treatment of multiple sclerosis using liquid chromatography with mass spectrometry detection. Curently, monoclonal IgG antibodies are used to treat multiple sclerosis. Attention will be focused to bio matrix modification methods in order to achieve rapid and simple sample treatment with the largest possible extraction yields. During the development of the method, the conditions of chromatographic separation will be tested: composition and flow of the mobile phase, type and temperature of the chromatographic column and volume of injection of the sample. Furthermore, mass spectrometer parameters will be tested: temperature and gas flow rates, ionic optics settings and identification and quantification MRM transitions will be found. The developed method will be validated according to the rules "European Medicines Agency" (EMA) for the validation of bioanalytical methods. Subsequently, the method will be used to analyze real blood serum samples of patients with multiple sclerosis and compared with the immunoassay ELISA method.
